ABSTRACT

Background Job burnout is an early mental health condition caused by job stress and contributes to many negative effects on work and life. Employees of research and development (R&D) enterprises are exposed to constant pressure from innovation, production speed and sales expansion, and they are prone to burnout symptoms if such factors are not under effective control. Objective To evaluate the current situation of job burnout among employees of R&D enterprises in Minhang District of Shanghai and explore its influencing factors. Methods During November to December 2021, a cross-sectional study was developed and a convenient sampling method was used to enroll employees from 7 R&D enterprises in Minhang District of Shanghai. On the basis of voluntary participation with informed consent, a survey was conducted by using a self-made questionnaire (collecting data about general demographic characteristics, occupational characteristics, behavior and lifestyle), the Chinese version of the Concise Occupational Stress Questionnaire, and the Chinese version of the Maslach Burnout Inventory-General Survey. Occupational stress and its dimensions (job demand, job control, and social support) were divided into high, medium, and low levels according to tertiles. The positive rate of job burnout was reported according to score categorization (<1.5 refers to no job burnout, ≥1.5 refers to job burnout, where ≥1.5 and <3.5 refer to mild and moderate job burnout, and ≥3.5 refers to severe job burnout). Potential influencing factors of job burnout were evaluated by using one-way ANOVA, chi-square test, forward stepwise regression, and non-conditional binary logistic regression (α=0.05, two-sided test). Results A total of 3153 subjects were enrolled and 3014 samples were included in the analysis, with a valid response rate of 95.6%. Among the included subjects, 888 (29.46%) reported no job burnout, 1775 (58.89%) reported mild to moderate job burnout, and 351 (11.64%) reported severe job burnout. The mean of total job burnout score was 2.17±1.12, and the dimentional mean scores were 2.78±1.61 for emotional exhaustion, 1.60±1.60 for cynicism, and 4.05±1.57 for diminished personal accomplishment. Varied categories of sex, age, marital status, working position, sleep status, job demand, job control, and social support groups of workers resulted in significant differences in job burnout score. Compared with the low job demand group, the positive rate of job burnout was elevated in the medium and high job demand groups; the risk of job burnout in the medium job demand group was 1.42 (95%CI: 1.04-1.94) times higher, and that in the high job demand group was 2.64 (95% CI : 2.17-3.22) times higher versus the low job demand group. The risk of job burnout in the medium job control group was 1.35 (95%CI: 1.06-1.72) times higher versus the low job control group. Compared with the low social support group, job burnout was less reported in the other groups, and the OR (95%CI) values of the medium and high social support groups were 0.41 (0.31-0.53) and 0.15 (0.12-0.19) respectively. Conclusion The rate of reporting positive job burnout in R&D enterprises is high, which deserves sufficient attention. Relieving work pressure, increasing job control and social support, and maintaining adequate sleep are helpful to reduce job burnout.

ABSTRACT

Background The tobacco epidemic as one of the most serious public health problems in the world contributes great harm to human health. It is urgent to develop tobacco control strategy. Civil servants' behaviors as a role model for society have a great influence on the rest of society. Thus, it is important to promote tobacco control program on helping civil servants to quit smoking. Objective To understand the smoking and second-hand smoking exposures status, the awareness of knowledge of tobacco hazards, and the attitudes to tobacco control policies of civil servants in Minhang District, Shanghai. Furthermore, to explore the related strategies and measures for tobacco control, to provide a basis for the development and revision of relevant intervention measures and policies. Methods A total of 20 government agencies with newly installed smoking-free programs from 10 subdistricts and towns in Minhang District, including Xinzhuang, Wujing, Maqiao, Qibao, Meilong, Hongqiao, Zhuanqiao, Jiangchuan, Gumei, and Xinhong were selected as study sites.A questionnaire survey was conducted to investigate all civil servants (n=801) of selected agencies. The questionnaire included general characteristics, smoking and second-hand smoking exposure status, knowledge of tobacco hazard, and attitudes to tobacco control policies. Results A total of 794 civil servants returned valid questionnaires. The awareness rate of tobacco hazards in the smoking group was lower than that in the non-smoking group (P<0.05). Relatively high smoking rates were reported in those being male (17.08%), age ≥51 years old (25.61%), divorced or widowed (12.50%), technical secondary school or below education level (22.81%), having more than 10 years of working years (11.57%), being willing to dissuade indoor smoking (24.54%), having no punishment or warning measures for violation of smoking regulations in workplaces (18.37%), and having no dedicated smoking spot at home (15.38%). The results of multiple logistic regression analysis showed that age 31-40 years old (OR=3.446, 95%CI: 1.236-9.609) and to avoid confronting indoor smoking (OR=3.686, 95%CI: 1.041-13.049) were risk factors for smoking in civil servants. Conclusion Civil servants aged 31-40 years old who deliberately avoid confronting indoor smoking behaviors are the key intervention population of smoking control in Minhang District, Shanghai.

ABSTRACT

Objective:To determine mechanisms for the role of high mobility group box-1 (HMGB1) to platinum based chemo-sensitivity in cervical cancer.Methods:Using Western blot,we examined the effect of cisplatin on expressions of LC3,Beclin1 and P62 in cervical cancer.After treated with autophagy inhibitor and/or cisplatin in HeLa and CaSki cells,cell counting kit-8 (CCK-8) assay was used to evaluate the chemo-sensitivity.The expression of LC3,Beclin1 and P62 were detected by Western blot.We established HeLashHMGB1,CaSki-shHMGB1 and HeLa-CTR,CaSki-CTR stable cell lines.CCK-8 assay was used to determine the chemosensitivity in these cell lines.Using Western blot,we examined the correlation among the expressions of HMGB1,LC3,Beclin1,and P62.Results:With the increase of cisplatin,the expression of LC3 and Beclin1 was increased,while the expression of P62 was decreased in HeLa and CaSki cell lines.Combination of cisplatin with autophagy inhibitor could increase the cellular sensitivity to cisplatin (P<0.05).The expression of HMGB1 was related to chemo-sensitivity and autophagy related protein in cervical cancer cells.HMGB1 was positively correlated with LC3 and Beclin1 while negatively correlated with P62 (P<0.05).Conclusion:HMGB 1 might play an important role in chemo-sensitivity via regulating autophagic activity.The combination of cisplatin with autophagy inhibitor might become a possible strategy for cervical cancer.HMGB1 might serve as a potential biomarker for predicting chemo-therapeutic responses.

ABSTRACT

Objective:To investigate effects of over-expression and suppression of HMGB1 on proliferation and invasion of endometrial carcinoma HEC-1A cell and underlying mechanisms.Methods:Over-expression or silence of HMGB 1 in HEC-1A cell lines were established by lentiviral vector containing HMGB 1 recombinant plasmid or by HMGB 1 shRNA,respectively.Cell counting kit-8,Transwell,and wound healing assay were used to analyze proliferation,invasion,and migration of HEC-1A cells,respectively.Western blot and reverse transcription-PCR (RT-PCR) were used to detect the expression of NF-κB,VEGF,and matrix metalloproteinase 2 (MMP2) in the cells.Results:Over-expression of HMGB1 promoted the proliferation,invasion,and migration of HEC1A cell,and up-regulated NF-κB,VEGF,and MMP2 expressions,while suppression of HMGB1 inhibited the proliferation,invasion,and migration of HEC-1A cells and down-regulated NF-κB,VEGF,and MMP2 expressions.Conclusion:HMGB1 plays an important role in proliferation,invasion,and migration of HEC1A cell via NF-κB/VEGF/MMP2 pathway.HMGB 1 might be a potential target for endometrial carcinoma therapy.

ABSTRACT

OBJECTIVE@#To determine the effects of p38 mitogen-activated protein kinase (MAPK) signal pathway after small hairpin RNA on inhibiting the expression of high mobility group box-1 (HMGB1), and to explore the mechanism responsible for the effects of HMGB1 on invasion and migration of endometrial carcinoma of uterus.
@*METHODS@#Based on RNA interference technology, we designed and synthetized HMGB1 small hairpin RNA (HMGB1 shRNA-1, HMGB1 shRNA-2, HMGB1 shRNA-3, HMGB1 shRNA-1), and then transfected HMGB1 shRNA plasmids into HEC-1A cells. The expression of genes and proteins was examined by RT-PCR and Western blot. The activity of HEC-1A cells was evaluated by the methyl thiazolyl tetrazolium (MTT) method.
@*RESULTS@#HMGB1 shRNA effectively inhibited the expression of the HMGB1 and p38MAPK in HEC-1A cells (P<0.05). Among the shRNAs, the inhibitory effect of HMGB1 shRNA-3 was the best (P<0.05). According to the results of MTT, the growth of HEC-1A cells was obviously inhibited after transfection of HEC-1A HMGB1 shRNA (P<0.05). 
@*CONCLUSION@#Downregulation of HMGB1 may effectively inhibit proliferation, invasion and migration of HEC-1A cells in endometrial carcinoma of uterus, which is dependent on p38MAPK signal pathway.

ABSTRACT

Objective To investigate the expression and clinical significance of DJ-1 protein in the endometrial carcinoma and delineate its functional roles in endometrial carcinoma progression.Methods Expression of DJ-1 was examined with immunohistochemical staining of 69 paraffin-embedded endometrial carcinoma samples who underwent surgery at Hunan province Tumor Hospital.Kaplan-Meier analysis was performed to describe the survival curve.Univariate and multivariate analyses were performed with COX proportional hazard model.Results (1) The expression of DJ-1 protein in endometrial carcinoma was significantly higher than that in normal endometrial tissues (P < 0.05).(2) There was positive correlation between the high expression of DJ-1 protein in endometrial carcinoma samples and the pathological grade,the International Federation of Gynecology and Obstetrics (FIGO) stage,depth of myometrial invasion,or lymph node metastasis (P < 0.05).(3) Kaplan-Meier analysis indicated that the cumulative 5-year survival rate in the low-DJ-1 expression group was 80.8%,whereas it was only 51.2% in the high-DJ-1 expression group (P < O.05).COX analysis indicated that the expression of DJ-1 was not the endometrial carcinoma independent prognostic factors for overall survival time (P > 0.05).Conclusions The high expression of DJ-1 protein in endometrial carcinoma samples demonstrated a significantly positive correlation with the pathological grade,FIGO stage,depth of myometrial invasion,and lymph node metastasis.However,DJ-1 is not enough to be independent prognostic factor for overall survival time.

ABSTRACT

Objective To investigate the efficacy and tolerability of estradiol (E2) gel with postoperative patients of moderately severe ovarian endometriosis cyst,who suffer from the side effect of GnRH agonist.Methods Sixty samples were selected with stage Ⅲ ~ Ⅳ ovarian endometriosis after conservative operation by the same operator at the Department of Gynecology,Xiangya Hospital,Central South University,between February 2013 and August 2014.All of the patients after surgeries were randomly divided into three groups:control group (n =20) with only goserelin injection after conservative surgery,treatment group A (n =20) who received estradiol gel,and treatment group B (n =20) who received tibolone as an add-back therapy.The serum E2,follicle stimulating hormone (FSH),luteinizing hormone (LH),hypoestrogenic symptoms,the incidence of uterine bleeding,and the recurrence after conservative operation were compared among three groups.Results Three months later aster treatment with goserelin,the serum level of E2 of three groups showed statistically significant difference (P < 0.05).Serum E2 level of treatment group A was not significantly higher than treatment group B (P > 0.05).The kupperman sore,the incidence of hot flashes sweating,tiring,and agrypnia of treatment groups A and B were significantly lower than the control group (P < 0.05).The incidence of uterine bleeding and recurrence dont differ sharply among three groups (P > 0.05).Conclusions GnRHa combined with estradiol gel can improve symptoms of postoperative patients,reduce side effects effectively,and avoid increase of the risk of vagina bleeding and relapse.

ABSTRACT

Objective To investigate the association between high-risk types of human papillomavirus (HR-HPV) load and the expression of nestin in different cervical lesions.Methods The hybridization capture Ⅱ (HC-Ⅱ) assay was used to test the HR-HPV load from 60 patients who were the first time to do the cervical cancer screening in Xiangya Hospital,and immunohistochemistry was used to detect the expression of nestin in those biopsy tissue samples.Results (1) The lgHPV level (logarithm of HR-HPV load) in the high level lesion group was higher than the low level one (P <0.05),and HR-HPV load was positively associated with the degree of cervical lesions (rs =0.269,P =0.037).(2) The expression of nestin in A group was weaker than groups B and C (H =7.271,22.843,P <0.01),and the expression of nestin in C group was stronger than B group (H =7.270,P <0.01),and the expression of nestin was positively associated with the degree of cervical lesion (rs =0.646,P =0.000).(3) The HR-HPV load was positively associated with the expression of nestin (P < 0.05).Conclusions The HR-HPV load and the expression of nestin are closely related to the cervical lesions,and the joint detection has a referential value for early prevention of cervical lesions and prediction of progress of cervical precancerous lesion.It might guide the early prevention and treatment of cervical cancers.

OBJECTIVE@#To investigate the effect of inhibiting high mobility group box-1 (HMGB1) gene expression on the invasion and migration of endometrial carcinoma HEC-1A cells by small hairpin RNA.@*METHODS@#Three specific recombinant plasmids of HMGB1 (pshRNA-1 /HMGB1, pshRNA-2 / HMGB1, and pshRNA-3/HMGB1) were transfected into the endometrial cancer cell lines HEC- 1A by lipofectamine (TM) 2000. The expression of HMGB1 mRNA and protein was decteted by RTPCR and Western blot. The invasion and migration abilities of transfected HEC-1A cells were evaluated using Transwell assay and wound healing assay.@*RESULTS@#RT-PCR and Western blot revealed that the expression of HMGB1 at both mRNA and protein levels was significantly inhibited by HMGB1-pshRNA targeting sequence 1, 2, and 3 (P<0.05), and the levels of 3 mRNAs in the transfection group were 0.192±0.006, 0.055±0.002, and 0.123±0.086, respectively, which were significantly lower than those in the Lipo group (0.268±0.008) and the HMGB1/p-NC group (0.270±0.004). The maximum inhibiton rates of the 3 mRNAs were 28.4%, 79.5%, and 54.1%. The levels of 3 HMGB1 proteins in the transfection group were 0.259±0.129, 0.032±0.002, and 0.104±0.007, significantly lower than those in the Lipo group (0.347±0.007) and the HMGB1/p-NC group (0.349±0.007), and the maximum inhibitory rates were 25.4%, 90.8%, and 70.0%. The transwell chamber assay and wound healing assay showed that the invasion and migration of HEC-1A cells were effectively suppressed by inhibiting HMGB1 expression (P<0.05).@*CONCLUSION@#pshRNA-HMGB1 can effectively inhibit HMGB1 expression at both mRNA and protein levels, and decrease the invasion and migration of endometrial cancer cells.

ABSTRACT

Objective To investigate the influence of CD146-siRNA on epithelial-mesenchymal transition in human ovarian cancer cell line A2780.Methods The influence of loss of CD146 on the mRNA and protein expressions of E-cadherin,vimentin,and fibronectin was evaluated by real-time polymerase chain reaction (RT-PCR) and Western blotting.Results Compared to non-transfected (control group) and those transfected with RNAi-NC (RNAi negative control group),the mRNA and protein expressions of Ecadherin were significantly increased in A2780 cell lines transfected with RNAi-CD146.The loss of CD146 expression down-regulated the mRNA and protein expression of vimentin and fibronectin(P < 0.05).Conclusions CD146-siRNA can suppress epithelial-mesenchymal transition in human ovarian cancer cell line A2780.

ABSTRACT

Objective: To investigate the effect of HMGB1 small interfering RNA (siRNA) on the proliferation, cell cycle and apoptosis of human endometrial cancer cell line HEC-1A, and itspossible molecular mechanism. Methods: Lentivirus vector with HMGB1 shRNA was constructed and infected the endometrial cancer cell line HEC-1A. After viral infection for 72 h, real time PCR and Western blot were performed to investigate HMGB1 mRNA and protein expression. The cell proliferation was determined with methyl thiazolyl tetrazolium (MTT) method. Flow cytometry was performed to analyze the cell cycle progression of propidium iodide (PI)-stained HEC-1A cells and the apoptotic rate of annexinV/PI-stained cells. Western blot was used to detect the protein expression of AKT, pAKT and CyclinD1. Results: Lentivirus vector with HMGB1 shRNA inhibited the mRNA (P<0.05) and protein (P<0.01) expression of HMGB1 in the cell line HEC-1A. The MTT assay demonstrated that HMGB1 knockdown signiifcantly reduced the cell proliferation. FCM results showed that HMGB1 knockdown significantly resulted in the disruption of the cell cycle at G0/G1 phase and the induction of apoptosis. hTe apoptotic rate was (17.89±0.23)%, (4.69±0.20)% and (4.62±0.17)% in the HMGB1 knockdown group, the blank group and the negative group respectively. There was signiifcance difference between the 3 groups (P<0.01). hTe protein expressions of pAKT and cyclinD1 were down-regulated atfer the HMGB1 knockdown for 72 h. Conclusion: Knockdown of HMGB1 expression can significantly inhibit the proliferation and induce the cell cycle arrest and apoptosis in the endometrial cancer cell line HEC-1A. PI3K/AKT pathway and down-regulation of the protein expression of cyclinD1 may be involved in its therapeutic mechanism.

Objective To investigate the expression of high mobility group protein box1 (HMGB1)in ovarian endometriosis tissue and to provide new idea for early diagnosis,treatment and prognosis of endometriosis.Methods The levels of HMGB1 in ectopic endometrium and eutopic endometrium of 69 patients with ovarian endometriosis and endometrium of 69 cases with non-endometrioses were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western-blotting.Results HMGB1 was expressed in all the three groups (ectopic endometrium eutopic endometrium endometrium with non-endometrioses).The relative expression level of HMGB1 mRNA was 0.54383 ± 0.0565,0.3016 ± 0.0614,and 0.1536 ±0.0444,respectively (F =377.923,P < 0.0001).The relative of HMGB1 protein was 0.83990 ±0.12869,0.40100 ±0.16425,and 0.19986 ± 0.08162,respectively (F =185.625,P =0.0000).The expression level of HMGB1 was related to clinical stages (t =10.28,P <0.01) ; however,it was not obviously related to age,the diameter of lump,and course of disease (P > 0.05).Conclusions Over expression of HMGB1 in ovary endometriosis indicates that HMGB1 plays a significant role in occurrence and development of ovary endometriosis.The expression of HMGB1 in endometriosis eutopic endometrium was much higher than that of non-endometrioses endometrium,which supports the eutopic endometrium determinism.The overexpression of HMGB1 was consistent with clinical stage,which proves that HMGB1 maybe be tightly associated with the severity of the disease and to likely become biomarker.

ObjectiveThe purpose of this study was to investigate the clinical significance of HMGB1 and Smac/DIABLO protein expression in epithelial ovarian cancer.MethodsImmunohistochemistry (IHC) and terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) staining were used to detect the protein expression of HMGB1,Smac/DIABLO and cell apoptosis in 45 epithelial ovarian cancers by tissue microarray.ResultsThe protein expression of HMGB1 in epithelial ovarian cancers was significant higher than benign cancers and normal ovarian tissues( P <0.05).The protein expression of HMGB1 was related to the lymph node metastasis,which had a negative correlation with the cell apoptosis index ( rs =-0.583,P =0.000).The protein expression of Smac/DIABLO in epithelial ovarian cancers was significantly lower than benign cancers and normal ovarian tissues( P <0.05) ; the protein expression of HMGB1 in epithelial ovarian cancers had a negative relationship with protein expression of Smac/DIABLO( rs =-0.40,P =0.006).ConclusionsThe over-expression of HMGB1 in epithelial ovarian cancers may play an important part in the metastasis of the epithelial ovarian cancers.The over-expression of HMGB1 and the low-expression of Smac/DIABLO may take part in the occurrence and development of the epithelial ovarian cancers through abnormal cell apoptosis.

ABSTRACT

Objective To investigate the expression of HMGB1 mRNA in endometrial carcinoma and its correlation with clinicopathological features of endometrial carcinoma. Methods Semi-quantitative RT-PCR method was used to detect the expression level of HMGB1 gene in 56 cases of endometrial carcinoma and 20 cases of normal endometrium. Results The expression of HMGB1 mRNA in endometrial carcinoma (0. 3512 ± 0. 0985 ) was significantly higher than that in normal endometrium (0. 2208 ± 0. 0170 ).There was a significant difference in the two groups( P <0. 05 ). Grouping by clinicopathological features,the expression of HMGB 1 mRNA in endometrial carcinoma had a correlation with clinical-surgical stage ( P<0. 05), metastasis of lymph nodes ( P < 0. 05), and depth of myometrial invasion ( P < 0. 05 ). Conclusion The high level of HMGB1 mRNA indicated that HMGB1 might play an essential role in the genesis, growth, invasion and metastasis of endometrial carcinoma.

Objective To study the causes of the uterus rupture in an outlying district and prevent measures.Methods 17 cases of the uterus rupture in Yongshun county hospital were retrospectively analysized.Results Among 17 cases of uterus rupture,the average antenatal care was 0 4 times every pregnant women,bleeding volume were 1100 ml,12 cases of shock(70 6%) ,5 cases forced uterectomy(29 4%), 16 cases of perinatal death(94%). According to stages of taking place : ⑴15 cases had taken place during laboring without appointing inpatient ,13 patients giving birth at home while 2 patients in hospital , average total stages of labor 29.4h, abnormal fetal positions 14 cases, including 3 cases of primipara and 12 cases of multipara. ⑵ 2 cases of spontaneous uterus rupture occurred during pregnancy who have with the cesarean section history performed in small clinic in the country side.Conclusions In an outlying district, main causes of uterus rupture:⑴ Ignored antenatal care. ⑵ Producing at home so that dystocia were not dealt with properly during stage of labor, another was women with prolong labor treated incorrectly . ⑶ The Spontaneous rupture during pregnancy were close related to the history of the classical cesarean section .To prevent the uterus rupture should be enhanced prenatal care and examination forced pregnant women delivery in hospital, standarding medical practice of performing operation, so doing could sharply decrease the pregnant uterus rupture in an outlying district.

ABSTRACT

Objective To investigate the cumulative organ damage of systemic lupus erythematosus (SLE) and It's affecting factors.Methods 162 cases of SLE patients were reviewed and evaluated in the cumulative organ damage of them . At the same time, It's affecting factors were analysed by multifactorial analysis.Results The cumulative organ damage of SLE was obviously correlated with the first time of treatment, treatment with CTX ,the numbers of recurrence rate and level of complements;but were not correlated with disease duration and cumulative dosage of corticosteroid.Conclusions The cumulative organ damage was one of the evaluating factors of SLE disease, and it's occurence and development were affected by multiple factors .So, the patients should be treated with hormone and control the beneficial factors to protect organs,such as,observation on complemets level,high dosage cyclophosphamide pulse treatment etc.

ABSTRACT

Objective To investigate the changes of disease activity and clinical significance in the course of treatment in patients with SLE.Method 286 cases of SLE were reviewed and compared the changes of SLEDAI scores in different disease duration.Results The SLEDAI scores of patients whose first treatment courses less than 1 month and 1 to 3 months were significantly lower than those patients whose were 4 to 6 months and more than 6 months. After treatment for 2 months to 3 years, the SLEDAI scores were not correlated with cumulated dosage of corticosteroids.Conclusions For the patients of short first treatment course, the treatment could relieve SLE disease activity rapidly and effectively to some extent; while for the patients whose first treatment courses were relatively long ,the relif of disease activity was relatively slow. After treatment for 2 to 3 months, the disease of SLE patients was more active than other periods, and it was inclined to produce visceral damage. As mentioned above ,we should pay attention to this phenomenon.
